Why do you want to do basic settings?
If you want. Then choose transmission, basic settings and basic settings there.
Car engine on, transmission on P, handbrake engaged and keep the brake pedal pressed for the entire process. After it’s completed, recycle ignition and choose fast clutch adaptation and go for a drive and try to use gears including reverse.
You will hear loud banging and clicking during basic settings. It’s normal. Also it’s important that you recycle ignition after basic settings, otherwise the gears won’t engage.
If you want. Then choose transmission, basic settings and basic settings there.

That seems to be a characteristic of that gearbox. I have diesel Tiguan with that same gearbox and I have the same effect. Basic setting makes it go away for a while and then it slowly creeps back.
The fast clutch adaptation is the final step in the adaptation process. It starts an adjustment process for both of the clutch packs to "optimize" the shifts in odd and even gears. That's why it needs you to drive for a bit using all gears after the clutch adaptation is started.
It is not mandatory. If you don't do it, it will adapt during normal drive. It will just take a bit longer than with the adaptation enabled.

Make sure you check the fluid level in the box as per RTFM. I’ve had three DQ500’s now slightly underfilled from the factory (about 3/4th of a liter short) and all three drivers noticed improved shifting afterwards. One of them already had an oil change once but that garage just added the same quantity of oil as the amount they drained, thus never solving that problem.
